　　細(xì)胞凍存和復(fù)蘇實(shí)驗(yàn)原理:細(xì)胞凍存及復(fù)蘇的基本原則是慢凍快融，實(shí)驗(yàn)證明這樣可以大限度的保存細(xì)胞活力。目前細(xì)胞凍存多采用甘油或二甲基亞砜作保護(hù)劑，這兩種物質(zhì)能提高細(xì)胞膜對(duì)水的通透性，加上緩慢冷凍可使細(xì)胞內(nèi)的水分滲出細(xì)胞外，減少細(xì)胞內(nèi)冰晶的形成，從而減少由于冰晶形成造成的細(xì)胞損傷。復(fù)蘇細(xì)胞應(yīng)采用快速融化的方法，這樣可以保證細(xì)胞外結(jié)晶在很短的時(shí)間內(nèi)即融化，避免由于緩慢融化使水分滲入細(xì)胞內(nèi)形成胞內(nèi)再結(jié)晶對(duì)細(xì)胞造成損傷。

　　細(xì)胞凍存和復(fù)蘇實(shí)驗(yàn)注意事項(xiàng):

　　1. 從增殖期到形成致密的單層細(xì)胞以前的培養(yǎng)細(xì)胞都可以用于凍存，但好為對(duì)數(shù)生長(zhǎng)期細(xì)胞。在凍存前**好換一次培養(yǎng)液。

　　2. 將凍存管放入液氮容器或從中取出時(shí)，要做好防護(hù)工作，以免**。

　　3. 凍存和復(fù)蘇好用新配制的培養(yǎng)液。

　　當(dāng)細(xì)胞冷到零度以下，可以產(chǎn)生以下變化：細(xì)胞器脫水，細(xì)胞中可溶性物質(zhì)濃度升高，并在細(xì)胞內(nèi)形成冰晶。

　　如果緩慢冷凍，可使細(xì)胞逐步脫水，細(xì)胞內(nèi)不致產(chǎn)生大的冰晶;相反，結(jié)晶就大，大結(jié)晶會(huì)造成細(xì)胞膜、綱胞器的損傷和破裂。復(fù)蘇過(guò)程應(yīng)快融，目的是防止小冰晶形成大冰晶，即冰晶的重結(jié)晶。
